Use of nuclepore filters for counting bacteria by fluorescence microscopy.
Polycarbonate Nuclepore filters are better than cellulose filters for the direct counting of bacteria because they have uniform pore size and a flat surface that retains all of the bacteria on top of the filter. Although cellulose filters also retain all of the bacteria, many are trapped inside the filter where they cannot be counted. Before use, the Nuclepore filters must be dyed with irgalan ...
متن کاملCarbon monoxide oxidation by methanogenic bacteria.
Different species of methanogenic bacteria growing on CO(2) and H(2) were shown to remove CO added to the gas phase. Rates up to 0.2 mumol of CO depleted/min per 10 ml of culture containing approximately 7 mg of cells (wet weight) were observed. Methanobacterium thermoautotrophicum was selected for further study based on its ability to grow rapidly on a completely mineral medium. This species u...
متن کاملIdentification of the mouse karyotype by quinacrine fluorescence, and tentative assignment of seven linkage groups.
A karyotype of the mitotic chromosomes of the house mouse has been prepared based upon quinacrine fluorescence patterns. All 19 pairs of autosomes and the X and Y chromosomes have been identified. Examination of the chromosomes of the following translocation stocks, T(11;?)1Ald, T(3;?)6Ca, T(2;9)138Ca, T(2;12)163H, and T(9;13)190Ca, have led to the tentative assignments of autosomal linkage gro...

متن کاملImproved technique for identification and enumeration of methanogenic bacterial colonies on roll tubes by epifluorescence microscopy.
Methanogenic fluorescent colonies can be clearly identified on roll tubes by using an epifluorescence microscope equipped with a x 2 objective. Methanogenic and nonmethanogenic colonies could be counted in roll tubes prepared from methanogenic enrichment cultures. Late-developing colonies appearing after 25 days of incubation were mainly methanogenic.
